Thymosin Beta-4 and Actin: A Primer for Cell Migration Research

Thymosin beta-4 is one of the cell's main actin-sequestering peptides — and a classic tool in migration assays.

Thymosin beta-4 is a 43-residue peptide that binds monomeric G-actin and regulates the pool available for filament assembly. That role places it at the heart of in-vitro cytoskeleton and migration research.

The actin-binding motif

A central LKKTET sequence is key to actin interaction. Synthetic sequences such as TB-500 allow researchers to study this behaviour with a defined, high-purity reference material.

Common migration assays

  • Scratch (wound-closure) assays imaged over time
  • Transwell / Boyden chamber migration
  • Live-cell imaging of lamellipodia formation
  • Actin polymerisation assays with pyrene-labelled actin

Controls

Use mitomycin C or serum starvation to separate migration from proliferation, and include vehicle controls on each plate.

Related reagents: TB-500 10 mg. Each batch ships with documented HPLC purity and a Certificate of Analysis on request.
