Aliquoting Peptides: How to Avoid Freeze-Thaw Damage

Every freeze-thaw cycle costs a little integrity. Aliquoting once saves a study's worth of variability.

Researcher aliquoting peptide solutions into small vials to avoid freeze-thaw cycles

Repeated freezing and thawing promotes aggregation, ice-interface denaturation and concentration changes from evaporation. The simplest fix is to divide a reconstituted peptide into single-use portions.

A simple aliquoting workflow

  1. Equilibrate the lyophilised vial to room temperature.
  2. Reconstitute aseptically and swirl gently until dissolved.
  3. Dispense single-experiment volumes into low-binding tubes.
  4. Label with compound, concentration, lot and date.
  5. Freeze quickly and store at −20 °C or below.

Choosing tubes

Low-protein-binding polypropylene minimises losses for hydrophobic and lipidated peptides. Avoid glass for very dilute solutions unless silanised.

Related reagents: BAC water and peptide reagents. Each batch ships with documented HPLC purity and a Certificate of Analysis on request.
